Zebrafish muscle (cross-section)
Description:

A 48 hour postfertilization zebrafish embryo expressing GFP labeled α-actin (fast-twitch muscle) was fixed and sectioned at 12 µm. Muscle fibers were additionally labeled with anti-myosin heavy chain monoclonal antibody, F59 followed by TRITC conjugated secondary antibody (red). The orange signal is the overlay of GFP and myosin immunofluorescence in fast-twitch muscle. The slow twitch muscle on the periphery is labeled only with the F59/TRITC (red). Nuclei are labeled with To-Pro-3 (blue).

Methods:

Wide-field fluorescence on a Nikon TE2000 inverted microscope using a 20x objective lens (N.A. 0.75). Images were processed with MetaVue software.
